Gonadorelin: what the research covers
Synthetic GnRH, identical to the hypothalamic decapeptide. The same molecule produces opposite outcomes depending on whether it arrives in pulses or continuously, and that is the whole of its pharmacology.
What Gonadorelin is
Gonadorelin is pGlu-His-Trp-Ser-Tyr-Gly-Leu-Arg-Pro-Gly-NH2, the ten-residue hormone released by the hypothalamus to drive the pituitary. It is not an analogue: the sequence is the native one, with the pyroglutamate at one end and the glycinamide at the other both present as in the natural molecule.
Its receptor, GnRHR, is a class-A G-protein-coupled receptor on pituitary gonadotrope cells, signalling through Gq/11, phospholipase C and intracellular calcium. The output is luteinising hormone and follicle-stimulating hormone.
Native gonadorelin is cleared within minutes, which is both its characteristic limitation and the reason it remains the reference compound: analogue series exist precisely because this molecule does not persist, and any claim about what a substitution achieves is measured against it.
The pulse paradox
Delivered in pulses, gonadorelin drives gonadotropin release. Delivered continuously, it desensitises and downregulates its own receptor, and gonadotropin output falls below baseline. The same agonist at the same receptor, and the direction of the effect depends on the temporal pattern of exposure.
That property is not a curiosity; it is the basis of an entire therapeutic class, and in the laboratory it makes gonadorelin the tool for studying how a receptor system encodes frequency rather than amplitude. An experiment on pulse frequency needs a ligand that clears between pulses, and a long-acting analogue cannot provide that.
It also sits in a well-defined position in a hierarchy that this catalogue happens to cover completely: kisspeptin above it driving the GnRH neurons, and the gonadotropins HCG and HMG below it acting at the gonad. Few axes can be interrogated at every level with material from one supplier.
How Amino Club supplies it
Supplied lyophilised in sealed vials as Gonadorelin, specified at 99% purity or better by HPLC with identity confirmed by mass spectrometry and released against the Certificate of Analysis for its own batch, as documented on the lab testing page.
Both termini are modified in ways that are easy to get wrong in synthesis: a free glutamate instead of pyroglutamate at one end, or a free acid instead of the amide at the other, each produces a molecule with substantially reduced activity and a mass differing by only a little. That is what the mass determination on the certificate is for. The dry powder is held below −18 °C; solutions are markedly less stable and are prepared in small volumes, kept cold and dark. Diluents are under reconstitution supplies.
