MGF: what the research covers

Mechano growth factor is not an analogue of anything. It is a splice variant of IGF-1 that muscle produces when it is mechanically loaded, and the confusion on that point runs through much of the available literature.

What MGF is

The IGF-1 gene is transcribed into more than one product. When skeletal muscle is subjected to mechanical load or damage, splicing shifts towards the IGF-1Ec isoform, whose carboxy-terminal E domain differs from the common form. The twenty-four residue peptide corresponding to that unique E domain is what is sold and studied as mechano growth factor.

This matters because the obvious assumption — that MGF is a modified or truncated IGF-1 — is wrong, and it leads to the wrong expectations. MGF is a distinct gene product with its own reported activity, and published work indicates at least part of that activity does not require IGF-1R at all.

In solution the native peptide is strikingly short-lived, measured in minutes rather than hours. That single property is the reason a pegylated version exists.

Local signal, not systemic hormone

The architecture is the interesting part. IGF-1 circulates and acts broadly; MGF is produced at the site of loading and acts there. The published work centres on satellite cell activation and proliferation, early repair signalling after mechanical injury, and the time course by which the Ec isoform rises and then falls as the common isoform takes over.

For a study design, the implication is that MGF is a poor choice where sustained exposure is required and a reasonable one where the question concerns an acute local signal. An experiment that needs the latter profile will often run MGF against PEG-MGF precisely to separate the effect of the signal from the effect of its duration.
