About Oxytocin Acetate

The sequence is Cys-Tyr-Ile-Gln-Asn-Cys-Pro-Leu-Gly-NH2, closed into a six-residue ring by a disulfide bridge between the two cysteines, with a three-residue tail. It differs from vasopressin at only two positions, which is why selectivity between the oxytocin receptor and the vasopressin V1a receptor is a recurring question in binding studies rather than a given.

The oxytocin receptor is a class-A G-protein-coupled receptor signalling principally through Gq/11, phospholipase C and intracellular calcium release, with reported Gi coupling in some systems. Supplying the peptide as an acetate salt rather than a trifluoroacetate one matters for cell-based work, because residual trifluoroacetate from purification can itself affect cultured cells and confound low-concentration assays.

What the research covers

In-vitro literature on oxytocin spans two fairly separate fields. In smooth-muscle physiology it is the classical contractile agonist in isolated uterine and mammary tissue preparations, and the bioassay against which receptor antagonists are characterised. In neuroscience it is studied as a central neuromodulator: receptor distribution and binding, effects on neuronal firing in slice preparations, and interaction with dopaminergic and serotonergic systems in animal models of social behaviour. Receptor-selectivity work against the vasopressin receptors runs through both.

Handling Oxytocin Acetate

Supplied lyophilised in a sealed vial. Oxytocin is one of the less stable small peptides in solution: the disulfide bridge is susceptible to reduction and exchange, and aqueous solutions lose potency measurably over days even when refrigerated. Laboratories generally store the dry powder below −18 °C, prepare solutions in small single-use volumes, keep them cold and dark, and avoid repeated freeze-thaw cycles. Allow the vial to reach room temperature before opening so that moisture does not condense onto the powder.
