ARA-290: what the research covers

An eleven-residue peptide taken from the inside of erythropoietin: the part that signals tissue protection, deliberately separated from the part that makes red blood cells.

What ARA-290 is

Erythropoietin does two unrelated things. One is familiar: it drives red blood cell production through the classical EPO receptor homodimer. The other is tissue protection, mediated by a different assembly — a heterocomplex of the EPO receptor with the common beta receptor, usually called the innate repair receptor.

The two activities map to different parts of the molecule. ARA-290 is an eleven amino acid sequence derived from helix B of erythropoietin, the face that engages the protective receptor, and it does not engage the erythropoietic one. The practical consequence is a compound that reproduces the tissue-protective arm of EPO signalling without raising haematocrit, which is what made the separation worth doing in the first place.

It is also known in the literature as cibinetide.

Why the receptor split is the whole story

For an experimentalist, ARA-290 is less interesting as a repair peptide than as a dissociation tool. If an effect of erythropoietin persists with ARA-290, that effect belongs to the innate repair receptor. If it disappears, it belonged to the classical receptor or to the haematological change downstream of it. Few compounds let a question be partitioned that cleanly.

Published work concentrates on neuropathic and small-fibre neuropathy models, on inflammatory signalling and macrophage phenotype, and on ischaemia-reperfusion injury in several tissues. There is also clinical trial literature, which is uncommon in this catalogue and which constrains the mechanism more tightly than cell work alone would.

Compared with the better-known repair peptides here — BPC-157 and TB-500, both of which act broadly and through mechanisms still under debate — ARA-290 has an unusually well-specified target.
