MK-677: what the research covers
An orally active, non-peptide ghrelin receptor agonist with a half-life measured in hours rather than minutes. What separates ibutamoren from the injectable secretagogues.
What MK-677 is
MK-677, also known as ibutamoren, is not a peptide. It is a spiropiperidine small molecule that acts as an agonist at the same GHSR-1a receptor the peptide secretagogues target. It is grouped with them because of what it does, not what it is made of.
Two properties set it apart. It is orally bioavailable, where the peptide secretagogues are not — a small molecule survives the gut, a pentapeptide does not. And its half-life is on the order of several hours rather than minutes, which produces a sustained elevation of growth hormone and IGF-1 rather than the discrete pulse that Ipamorelin or GHRP-2 give.
Why the kinetics change the question
Growth hormone is secreted in pulses, and a good deal of its downstream signalling appears to depend on that pattern rather than on total exposure. A compound that flattens the pattern into a sustained elevation is therefore testing a different hypothesis from one that amplifies individual pulses, even where both are described as raising growth hormone.
That makes MK-677 the appropriate tool for work on chronic elevation and on IGF-1 as a downstream marker, and the wrong tool for work on pulsatility. It also means the reported observations that accompany sustained GHSR-1a agonism — effects on appetite signalling, on fluid balance and on insulin sensitivity — are part of the compound's profile rather than incidental to it, and any study design needs to account for them.
