CJC-1295: what the research covers

A modified GHRH analogue supplied in two forms whose half-lives differ by orders of magnitude. What CJC-1295 is, what the DAC changes, and how it is supplied.

What CJC-1295 is

CJC-1295 is a synthetic analogue of growth hormone releasing hormone, built on the first 29 residues of the natural sequence — the shortest fragment that retains full activity at the GHRH receptor. Four amino acid substitutions are made to that fragment, principally to resist cleavage by dipeptidyl peptidase-4, which degrades native GHRH within minutes.

It acts at the GHRH receptor on pituitary somatotrophs, amplifying the endogenous pulse rather than replacing it. That is the mechanistic difference between this class and recombinant HGH: the pituitary remains in the loop, and feedback regulation is preserved.

What the DAC changes

The two forms in the catalogue are the same core peptide. CJC-1295 with DAC carries a drug affinity complex — a maleimidopropionic acid group that forms a covalent bond with serum albumin after administration. Because albumin circulates with a half-life measured in weeks, the bound peptide persists far longer than the unmodified form, whose half-life is on the order of half an hour.

This is not a potency difference, it is a kinetic one, and it changes the shape of the experiment. The DAC form produces a sustained elevation; the form without DAC — often called modified GRF(1-29) — produces a discrete pulse that decays. Work concerned with pulsatility, or with pairing a GHRH analogue to a secretagogue so the two peaks coincide, requires the non-DAC form.
