About FOXO4-DRI

FOXO4-DRI is a retro-inverso peptide: built from D-amino acids in reversed sequence order, which produces a molecule presenting a similar side-chain arrangement to the natural L-peptide while being almost entirely resistant to proteolysis. That design is the reason it survives long enough to act.

Its target is the interaction between FOXO4 and p53. In senescent cells, FOXO4 sequesters p53 in the nucleus and prevents it from initiating apoptosis; disrupting that interaction is intended to release p53 and allow the senescent cell to die, while leaving healthy cells unaffected.

What the research covers

Work covers FOXO4-p53 binding disruption, apoptosis selectivity between senescent and non-senescent cells, and senescence markers such as p16, p21 and SA-beta-galactosidase. Clearance in ageing and chemotherapy-induced senescence models is the downstream endpoint.

Selectivity is the entire question. A compound that releases p53 indiscriminately is not a senolytic, so studies without a non-senescent comparator are not testing the property that matters. The longevity research peptides hub sets out the four separate hypotheses this group covers.

Handling FOXO4-DRI

Supplied lyophilised. The D-amino acid backbone makes this substantially more stable against proteolysis than any L-peptide of comparable length, which is an advantage in long cell-culture experiments where peptidase activity would otherwise degrade the agent.

Identity assurance rests on synthesis route rather than on mass spectrometry: the all-L sequence has the same mass and would be rapidly degraded and largely inactive, so stereochemistry cannot be confirmed by mass alone.
