Selank: what the research covers

An analogue of tuftsin, a four-residue immune peptide, extended by the same stabilising motif used in Semax. Its literature runs through anxiety rather than cognition.

What Selank is

Tuftsin is a tetrapeptide — Thr-Lys-Pro-Arg — released from the heavy chain of immunoglobulin G, with a well-documented role in stimulating macrophage and neutrophil function. Selank is a tuftsin analogue extended by Pro-Gly-Pro, the same protease-resistant motif used to stabilise Semax, giving the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro.

The two compounds came out of the same Russian research programme and share a design philosophy, but not a parent molecule and not a reported mechanism. Semax derives from a pituitary hormone fragment; Selank derives from an immune peptide. That difference shows up in what each one is studied for.

Anxiolytic without the receptor

The reported profile is anxiolytic, and the mechanistically interesting part is what is absent: Selank does not bind the benzodiazepine site on the GABA-A receptor. Published work instead describes modulation of GABA-A subunit expression, effects on serotonin turnover, and influence on the enkephalin system via inhibition of enkephalin-degrading enzymes — indirect routes rather than direct channel modulation.

That matters for a study design because the usual liabilities of benzodiazepine-site agonists — sedation, tolerance, dependence, withdrawal — are consequences of that binding, and a compound reaching an anxiolytic readout without it is pharmacologically a different thing. Whether the indirect mechanisms are sufficient to account for the reported effects remains an open question in the literature.

The immune strand has not disappeared either: Selank retains reported effects on cytokine balance and interleukin expression, inherited from its tuftsin parentage, and some authors treat the behavioural and immune findings as connected rather than separate.

The sequence is strongly basic, with lysine and arginine side chains, so it adsorbs to glass and ordinary plastic more readily than a neutral peptide — a consideration at low working concentrations. The powder is stored below −18 °C, solutions prepared in small volumes and kept cold. Diluents are under reconstitution supplies.
